Tau-381, also known as the 1N3R isoform of the microtubule-associated protein tau (MAPT), is a naturally occurring variant composed of 381 amino acids. It includes one N-terminal insert and three microtubule-binding repeat domains, contributing to its role in microtubule stabilization and neuronal plasticity. This isoform is particularly relevant in tauopathies characterized by selective 3R tau aggregation, such as Pick’s disease. Under pathological conditions, Tau-381 can misfold and assemble into β-sheet-rich fibrillar aggregates. Pre-formed fibrils (PFFs) generated from wild-type Tau-381 replicate key structural and biochemical features of tau aggregates found in human neurodegenerative diseases. These fibrils efficiently seed endogenous tau aggregation, promote neurofibrillary tangle formation, and drive neurodegenerative processes including synaptic dysfunction and neuronal loss. Tau-381 PFFs are widely used in experimental models to study tau propagation, misfolding, and toxicity. Their reproducibility and disease relevance make them ideal for investigating prion-like transmission, isoform-specific pathology, and therapeutic intervention strategies. They also serve as robust platforms for screening compounds that inhibit tau aggregation, enhance clearance, or restore neuronal function. By mimicking the core pathological features of 3R tau-driven neurodegeneration, Tau-381 wild-type PFFs accelerate translational research and therapeutic development for tauopathies such as Pick’s disease and Alzheimer’s disease. |
