Tau-412, also known as the 1N4R isoform of the microtubule-associated protein tau (MAPT), is a naturally occurring variant composed of 412 amino acids. It includes one N-terminal insert and four microtubule-binding repeat domains, contributing to its role in stabilizing microtubules and maintaining neuronal polarity and axonal integrity. Under pathological conditions, Tau-412 can misfold and aggregate into β-sheet-rich fibrillar structures. Pre-formed fibrils (PFFs) generated from wild-type Tau-412 replicate key features of tau aggregates found in tauopathies such as Alzheimer’s disease, progressive supranuclear palsy, and corticobasal degeneration. These fibrils are capable of seeding endogenous tau aggregation, promoting the formation of neurofibrillary tangles and driving neurodegenerative cascades. Tau-412 PFFs are widely used in experimental models to study the mechanisms of tau propagation, misfolding, and toxicity. Their reproducibility and disease relevance make them ideal for investigating prion-like transmission, synaptic dysfunction, and neuronal loss. Additionally, they serve as robust platforms for screening therapeutic agents aimed at inhibiting tau aggregation, enhancing clearance, and restoring neuronal function. In vitro seeding activity of human Tau-412 (1N4R) monomer in ThT assay. Tau 1N4R pre-formed fibrils seed fibril formation of human Tau-412 (1N4R) monomers over 72 hours. Reactions (100uL) shaken at 600 rpm in Greiner-Bio 96 Well Non-Binding Cell Culture Microplates, Black (Greiner-Bio Catalog #655900) at 37oC in the presence of 25 uM ThT and 10 uM heparin and read with an XPS Microplate Reader set at 450nmex/485nmem.
Sedimentation assay on E. coli expressed hTau-412 (1N4R) Pre-formed Fibrils. Samples were spun down at 15,000 x g, washed, and then spun down again. Fibril samples are prepared in denaturing conditions prior to running on the gel. SDS-PAGE analysis on a 12% Bis-Tris gel shows that the majority of the fibril prep shifts to insoluble post-fibrillization of monomers.
