Tau-441, also known as the 2N4R isoform of the microtubule-associated protein tau (MAPT), is a full-length wild-type variant comprising 441 amino acids. It includes two N-terminal inserts and four microtubule-binding repeats, making it the longest isoform expressed in the adult human brain. Tau-441 plays a critical role in stabilizing neuronal microtubules and maintaining axonal integrity. However, under pathological conditions, Tau-441 can misfold and aggregate into β-sheet-rich fibrillar structures, known as pre-formed fibrils (PFFs). These Tau-441 PFFs are increasingly recognized as key drivers of tauopathy progression, including Alzheimer’s disease, frontotemporal dementia, and other neurodegenerative disorders. When introduced into cellular or animal models, Tau-441 PFFs seed endogenous tau aggregation, recapitulating hallmark features of disease pathology such as neurofibrillary tangles, synaptic dysfunction, and neuronal loss. Their ability to propagate pathology in a prion-like manner makes them a powerful tool for studying disease mechanisms and testing therapeutic interventions. Tau-441 PFFs serve as a reproducible and physiologically relevant model for investigating tau aggregation, neurotoxicity, and intercellular transmission. Their use in preclinical research accelerates the development of targeted therapies aimed at halting or reversing tau-driven neurodegeneration. Tau 2N4R oligomers (catalog # SPR-497) and fibrils (catalog # SPR-498) show a dose-dependent toxicity to primary rat hippocampal neurons, but not monomers (catalog # SPR-496). Survival of rat primary hippocampal neurons 14 days after treatment with different concentrations of (A) monomers, (B) oligomers or (C) fibrils quantified by MAP2 positive neurons and expressed as a percentage of control. Fibrils were initially sonicated in a Bioruptor. Test conditions were run in the same plate as untreated control, which consisted of buffer without Tau protein. Data expressed as mean +/- s.e.m. (n=6). A global analysis of the data was performed using a one-way ANOVA followed by Dunnett’s test; * p < 0.05, ** p < 0.01, *** p < 0.0002, **** p < 0.0001; stats vs control.
