| Scientific Background |
Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and function. The Tau-441 isoform (2N4R), the longest variant expressed in the adult human brain, contains two N-terminal inserts and four microtubule-binding repeats, making it a key reference in tauopathy research. Under pathological conditions, wild-type Tau can misfold and aggregate into β-sheet-rich fibrils, contributing to the formation of neurofibrillary tangles observed in Alzheimer’s disease and other tauopathies. Pre-formed fibrils (PFFs) generated from wild-type Tau-441 replicate early aggregation events and provide a physiologically relevant model for studying the mechanisms of Tau seeding, propagation, and neurotoxicity. Tau-441 wild-type PFFs are widely used in cellular and animal models to investigate the prion-like spread of Tau pathology, synaptic dysfunction, and neuronal degeneration. Their reproducibility and structural fidelity make them ideal for evaluating therapeutic strategies aimed at preventing aggregation, promoting clearance, and interrupting intercellular transmission. As a foundational tool in neurodegenerative disease research, Tau-441 wild-type PFFs enable high-resolution studies of Tau biology and support the development of targeted interventions to halt or reverse disease progression. |

Thioflavin T is a fluorescent dye that binds to beta sheet-rich structures, such as those in tau fibrils. Upon binding, the emission spectrum of the dye experiences a red-shift and increased fluorescence intensity. Thioflavin T emission curves show increased fluorescence (correlated to tau aggregation) over time when tau wild-type monomers (SPR-479) are combined with tau wild-type fibrils (SPR-480).
