Scientific Background | Bafilomycin A1 is a macrolide antibiotic that selectively inhibits vacuolar-type H⁺-ATPases (V-ATPases), disrupting lysosomal acidification and autophagic flux. In neuroscience, Bafilomycin A1 is widely used to study autophagy, a critical process for neuronal health and protein clearance. By blocking autophagosome-lysosome fusion or lysosomal degradation, Bafilomycin A1 allows researchers to assess autophagic activity and flux in neuronal cells. Impaired autophagy is a common feature in neurodegenerative diseases such as Alzheimer’s, Parkinson’s, and ALS. Bafilomycin A1 is instrumental in elucidating the role of lysosomal dysfunction in these conditions and in evaluating therapeutic strategies aimed at restoring autophagic balance. |

We ordered this inhibitor and utilized 100 nm to pre-treat differentiated THP-1 cells for 1 h before incubating with autophagy inducer. We showed that this inhibitor can efficiently inhibit the autophagy flux by measuring the level of LC3-II. We recommend it.
