Scientific Background SP600125 is a selective, reversible inhibitor of c-Jun N-terminal kinases (JNK1, JNK2, and JNK3), with IC50 values ranging from 40 to 90 nM. JNK signaling is implicated in neuronal apoptosis, neuroinflammation, and synaptic dysfunction—hallmarks of neurodegenerative diseases such as Alzheimer’s and Parkinson’s. SP600125 is frequently used to dissect the role of JNK in neuronal cell death pathways and to evaluate neuroprotective strategies. Although it exhibits reduced selectivity under certain conditions, its in vivo activity and minimal off-target effects on ERK2 and p38β make it a valuable tool for studying stress-activated kinase signaling in the brain. SP600125 has also shown protective effects in models of ischemia-reperfusion injury, supporting its relevance in stroke and neurovascular research.
