Scientific Background Wortmannin is a fungal metabolite that irreversibly inhibits phosphatidylinositol 3-kinase (PI3K), a key regulator of intracellular signaling pathways involved in growth, metabolism, and autophagy. In neuroscience, Wortmannin is used to investigate the PI3K/Akt/mTOR pathway, which plays a central role in neuronal survival, synaptic plasticity, and neuroinflammation. By inhibiting PI3K, Wortmannin suppresses autophagy and disrupts membrane trafficking—processes essential for neuronal maintenance and function. Its application in neurodegenerative disease models helps elucidate the impact of impaired PI3K signaling on protein aggregation, mitochondrial dysfunction, and cell death. Wortmannin is also used to study insulin signaling in the brain, relevant to Alzheimer’s disease and metabolic-cognitive disorders.
